In phage display, the antigen binding regions of antibody heavy and light chain genes are cloned and expressed as fusion proteins on the surface of bacteriophage. These phage displayed antibody fragments are used in selections for binding to immobilised/soluble antigens. Bound antigen-antibody complexes are eluted and enriched by bacterial infection. By repeated rounds of selection, an amplified antigen-binding phage antibody population is generated. This pool is screened for individual positive phage binders using biochemical and biological assays and unique clones are isolated based on DNA profiling.
