The facility offers selection and screening of antibody or peptide libraries against a wide range of targets and generation of positive hits in a number of different formats
Single chain Fv fragments (scFvs) – The variable heavy and light chain domains of an antibody which participate in antigen binding are linked together by a flexible linker for conformational stability and solubility. These fragments are displayed on the surface of bacteriophage and selected for antigen binding. Positive phage hits will be reformatted and expressed as,
Soluble antibody fragments (scAbs) – Single chain Fv (scFv) linked to the constant kappa light chain of a human antibody for stabilisation, purification and detection purposes. Molecular mass of scAb is normally around 40 kDa.
Monoclonal Fabs – The fragment antigen binding (Fab) format consists of the variable and one constant chain of each of the heavy and light chain of an antibody. Fab version weighs around 50-55 kDa.
Monoclonal IgGs -Positive antibody fragments will be reformatted into full mouse/ human IgGs (mAbs) and expressed transiently in mammalian cell expression system. Alternatively, stable cell lines for these clones will be generated and antibodies will be expressed in mammalian cell system.
Biochemical and cell-based assays will be optimised in all the above cases with significant input from the academic partner.
