My main research interests are related to two key physiological processes in salmonid fish, namely the control of protein deposition and immune responses. A major research gap is the relation between metabolism and immune competence in fish and the related expression of genes.

Protein metabolism:

The rate of protein deposition during growth is a balance between protein synthesis and degradation. These two processes are under tight control and evidence suggests that in fish the control of degradation may be key to efficient protein growth. There are two main cellular pathways that can degrade cellular proteins, the energy dependant ubiquitin proteasome pathway and the lysosomal degradation of proteins. Both theses processes are tightly regulated and their regulation in fish may be very different to what is known about mammals, as ecto and endothermic animals have different strategies for controlling protein mobilization. To address these questions I have used a combination of biochemical, microarray and proteomics approaches.

Immune responses:

The coordinated response of a fish to either a pathogen or vaccine requires the correct identification of the foreign material. Signalling pathways result in gene expression changes and the fish can then mount the desired response to eliminate the disease or have protection by the vaccination. We have developed a 17,000 gene microarray (see link to salmon TRAITS) from which we have identified gene expression profile changes for both bacterial infection and specific response to recombinant fish cytokines.  More recently we have moved to use custom designed Agilent 4 x 44K Atlantic salmon and rainbow trout microarray slides.

Molecular basis of improved feed quality for aquaculture 2008-2011, PhD CASE Studentship, Co funded with BioMar

The replacement of fish meal and fish oils using vegetable proteins and vegetable oils is hampered by "antinutritional factors" that can be co purified, leading to metabolic dysfunction in the fish.  This results in undesirable affects including reduced digestibility, reduced food conversion, gut inflammation, altered immune function and reduction in growth.  This project will examine the resulting gene expression modifications in fish fed alternate diets, and help identify key molecular markers which will assist in the selection of new diets regarding performance and health of the fish. Student: Luca Tacchi.

IMAQUANIM   Improved immunity of aquacultured animals (EU 2006-2011)

EU funded project: Based on important disease models, this project concerns development of a technological knowledge platform for a future improved immunity to infectious pathogens in the major aquacultured species in Europe (Atlantic salmon, rainbow trout, sea bream, sea bass, carp, mussel and oyster). Focus will be put on use of vaccines, immuno-stimulants, immuno-diagnostic surveillance as well as markers for selection of the most immuno- competent individuals.

Evolutionary dynamics of major histocompatibility genes in Arctic charr,  

NERC 2007-2010, PI Dr W.C Jordan Institute of Zoology, London.

 Arctic charr (Savelinus alpinus), in common with many salmonid species, show great variability within and across localities. A number of morphotypes of charr coexist in various freshwater systems and differ in characteristics such as body size and spawning coloration, external and internal morphology and parasite fauna. Differences in parasite faunas are likely to create differences in the types of natural selection experienced by different morphs and subsequent differentiation at loci involved in the immune response. The highly polymorphic major histocompatibility (MH) genes are likely targets for such selection. MH class I and class II molecules are instrumental in eliciting an immune response, which results in elimination or control of the pathogen. We will isolate and characterize the MH class I and class II genes from Arctic charr and use the cDNA sequence information to genotype charr populations from different localities.
