1) Molecular mechanisms of fish antiviral defence.

2) The role of Leucocytes in antigen presentation.

Current Research Projects

Development of leucocyte cell lines for immunological research in teleost fish (Funded by NC3R, 2009-2011)

Whilst many fish cell lines have been developed over the years, and are widely used in many aspects of biological studies in virology, physiology and toxicology, most have a fibroblast or epithelial cell phenotype. Cytokines are important growth factors for leucocyte development and differentiation. The promgramme is focused on establishing a number of rainbow trout leucocyte cell lines, using cytokines as growth factors, and on phenotyping the cells that grow in response to these factors, to enable their subsequent use by the research community. These cell lines will provide a permanent platform for basic research of the fish immune system and have potential application for testing of aquaculture drugs/reagents to complement the use of live fish, and to evaluate the impact of chemicals/pollutants or other factors in the environment that may impact on fish health and welfare. Characterising immune functions of chemokines in rainbow trout (Funded by the Royal Society of Edinburgh and the Leverhulme Trust Visiting Scholarship, 2009-2010)

Chemokines are a large family of soluble peptides directing cell migration in vertebrates. They play an important role in leucocyte trafficking and activation of the immune system. Chemokines are small in size, typically 8-10 kDa and can be classified into two major subfamilies, CXC and CC, based on the distinctive cysteine patterns at the N terminal region. As in mammals, fish appear to have a large repertoire of chemokines; with many having no apparent homology to mammalian molecules. This project is to focus on phylogenetic analysis and functional characterisation of trout chemokines, and on evaluation of the impact of bacteria and virus derived compounds on their gene expression.
